Pineal-derived melatonin is synthesized from tryptophan, which is first converted into 5-hydroxytrophan by tryptophan hydroxylase and then decarboxylated into serotonin (5-HT). Two additional steps transform 5-HT into melatonin, including the rate-limiting step catalyzed by the enzyme aralkylamineN-acetyltransferase (AANAT) to yieldN-acetylserotonin, followed by the transfer of a methyl group from S-adenosyl methionine to the 5-hydroxy group ofN-acetylserotonin by hydroxyindole-O-methyltransferase (HIOMT).Ref:Ronald M.
